Although they both give you tons of customization options from the number of home screens available (1-7), a customizable dock, and the options of a 2D or 3D (ie nexus one on froyo) app tray, the features added by the Plus version is well worth it (not to mention supporting the developer so the new features will continue to come).
The Plus version is set at $2.99 for early adopters and is well worth every penny. However, you will not find the Plus version in the Market because the developer does not live in an area that supports paid apps. You can get the Plus version by downloading and installing the APK directly, but I recomend downloading the free version from the Market first. This not only lets you get a feel for LauncherPro, but if you try to add a Plus only feature, it will also give you the option to purchase the Plus version. Doing it this way will allow you to be notified via the Market of updates to LauncherPro. If you download the APK directly, you will have to check back to ensure you have the latest and greatest version.
As mentioned before, the dock is completly customizable, allowing you the choice of one dock or up to 3 scrollable docks, each with it's own customizable shortcuts. Each item in the dock can be set using the default icons, LauncherPro Icons, or a custom icon you create.

I have this running on both my Nexus One (2.2) and my Samsung Vibrant (Galaxy S 2.1) and never had any issues with it bogging down the phone or any FC's caused by LauncherPro. For anyone looking to ditch their carrier tweaked UI's, such as the MotoBlur or TouchWiz, or just looking to spice up your stock Android device, this is a great launcher alternative and is constantly evolving.
